Pengaruh Perbedaan Konsentrasi Lactobacillus plantarum Terhadap Perubahan Kadar Kolesterol Total Pada Tikus Putih

Abstract

Kebiasaan mengkonsumsi makanan yang mengandung lemak dan gula tinggi akan menyebabkan obesitas, yang pada akhirnya akan menimbulkan sindrom metabolik. Desain penelitian ini menggunakan rancangan penelitian post test only control group, dengan menggunakan sampel rattus norvegicus galur wistar berwarna putih, berjenis kelamin jantan, dewasa berusia 3-4 bulan, dengan berat badan 150-200 gram, dan dengan kondisi fisik sehat yang digunakan sebagai hewan model kolesterol tinggi dengan diberi pakan tinggi lemak, yang kemudian diberi bakteri probiotik Lactobacillus plantarum. Variabel bebas pada penelitian ini adalah Lactobacillus plantarum, sedangkan variabel tergantung adalah kadar kolesterol total dan kolesterol LDL. Pemeriksaan kadar kolesterol total dan kolesterol LDL dengan metode enzimatik kolorimetri dengan prinsip COD-PAP dilakukan pada akhir penelitian. Tujuan penelitian ini adalah untuk mengetahui dosis optimum probiotik yang dapat menurunkan kadar kolesterol tinggi dalam darah. Analisis data menggunakan one way Anova, dimana hasil yang didapat menunjukkan tidak adanya perbedaan yang signifikan pada pemberian bakteri Lactobacillus plantarum dengan konsentrasi 5x10⁸CFU dibandingkan konsentrasi 5x10⁹CFU. Oleh karena itu, pemberian Lactobacillus plantarum dengan konsentrasi sebesar 5x10⁸CFU merupakan dosis optimum yang dapat menurunkan kadar total kolesterol dan LDL kolesterol dalam darah.
